Abstract | ||
RAPD marker used as one of PCR techniques to determine the genetic relationship, distance and DNA fingerprint of the parents involved in a diallel cross and their F1 hybrids .The DNA isolated from leaves of these genotypes, the amount of DNA ranged between ( 943-2942) micrograms per 0.5 g of plant leaves and purity ranged from (1.67 to 1.94), then conducted the PCR reactions and samples deported on agarose gel. RAPD reactions studied using 33 primers , 18 primer haven't any binding sites while 15 primer showed 84 band 3of them had general main bands and 71 was Polymorphic band.Genetic distance results showed that the lowest genetic distance (0.172)was between the parent IK8(6) and their hybrid with the parent IK58(1) while the farest genetic distance (0.927), between parent IK58(1) and their hybrid with parent ZP-301(3). DNA fingerprint have been identified through the unique band emergence for parents (IK-58, un44052, ZP-301and R-153) and some of their hybrids.The analysis of the genetic relationship (dendrogram) showed that there are (6) major groups, the first contain two subgroup, the first included both parents (IK-85) and (Un44052) which met with the second one (R-153). Second main group split into two subgroup the first one involving of two parent (ZP-301) and (ZP-707) and the second included (Un44052 × ZP-707) and (ZP-301 × IK-8) meet with them the hybrid (IK-85 × R-153), the second subgroup included the hybrids ( IK-85xZP-707) and (ZP-301 x Un44052) meet with them the hybrid (ZP-707 × R-153).. The third main group involving all of IK -8 , (IK-85 × IK-8) , (IK-851 × Un44052) , (Un44052 × IK-8) and (Un44052 × R153), The fourth main group included the hybrids (ZP-301 × R153) and (ZP-301 X Zp-707). The fifth main grope included the tow hybrids(IK-58 X ZP-301)and IK-58 X R-153) ,While the sixth include the hybrid (ZP-707 X IK-8) which take its own path to met with all other major groups at the end of the genetic line . | ||
