Abstract | ||
This study aimed to phenotypic detection of some virulence factors of uropathogenic Escherichia coli (UPEC) associated with urinary tract infection. A total of 150 urine sample have been collected from patient suffering from UTI whom admitted to AL- Najaf AL-Ashraf hospitals from both gender, involved 107 urine samples for female with aged group range between 14-50 years and 43 urine samples from male with age group range between 8-45 years. The results of microscopic, phenotypic and biochemical test showed that only 30 isolates were belong to UPEC in which 24 (22.42%) isolates from female and 6(13.95%) isolates from male. PCR technique has been used to detect the serotype of UPEC by using O15 and O25 serotype gene and the results of gel electrophoresis showed that only 5(16.66%) isolates were belong to O15 by appearing of amplicon with 183 bp while 6(20%) isolates were belong to O25 with 230 bp of amplicon . Phenotypic detection of some virulence factors in UPEC has been carried out. The results revealed that all isolates (100%) were biofilm formation in which the range of optical density (OD) of biofilm formation isolates were 0.12 – 0.4 at 640 nm wave length measured by spectrophotometer. Also the ability of UPEC to produce alkaline phosphotase has been detected by measuring the diameter of phospholytic zone and the results showed that all isolates were alkaline phosphotase producer with 10.33-21 mm phospholytic zone. On the other hand the results indicated that all isolates were capsule producer while they have not the ability to produce protease | ||
